"採樣大小：\r\n儘可能採取足夠量之組織，但以不破壞實體標本為主。一般魚體小於10cm者，採取約長寬高各約3mm左右等量之組織；體長介於10-20cm者，採取長寬高各約5mm左右等量之組織；體長大於20cm者，可採取長寬高各約1cm左右等量之組織。\r\n\r\n採樣份數：\r\n若組織體積足夠，分為三份，一份以酒精保存，兩份以液態氮保存，其中一份置於中研院主庫，一份置於動物園備份庫，同個體者標號相同。\r\n\r\n採樣部位：\r\n一般以特徵較少的尾柄肌肉為主，採取組織時請避開該類群重要之鑑定形態特徵，保留實體標本進行形態分類所需要之資訊。若尾柄有重要特徵，請以背部肌肉為主，採取組織時應避開測線，並請盡量以「ㄇ」字形切開表皮後採取組織，儘可能保留表皮及鱗片等構造。採樣時盡量避免採取表皮，以免體外微生物或黏液等污染。此外也應避免消化道，以避免食物或消化道微生物之污染。若採取體內器官之組織，可採取較少作為鑑定特徵之肝臟，粒線體數量亦較多，但請特別小心不可破壞膽囊，因膽汁會破壞採取之組織。若魚體極度微小或脆弱，可剪下較不完整之一側或右側的偶鰭（胸鰭、腹鰭），或翻開鰓蓋剪下小塊鰓絲，但儘可能保留重要特徵。採樣使用之器具剪刀等，請在處理一個個體後完成清理及消毒程序，才能處理下一個個體，以避免污染，可以酒精燈火烤進行之。", Temperature）意即近表面之海水溫度。這裡指的「表面」，因測量方式不同而異。紅外線衛星輻射儀僅能間接測量海水表層約10微米厚度之溫度（因紅外線輻射從此薄層放出）；微波測量則可達表層下約1毫米；固定或是漂浮浮標上的溫度計則能測量到特定深度的溫度，如：海平面下1公尺深處；一般船隻測量，由發動機進水口測得，則可能介於海水表層至20公尺之深度不等處。因此，會因測量工具不同而測得不同深度之「海水表面溫度」。\r\n\r\n海水表面溫度對於氣候和天氣有著舉足輕重的地位。舉例來說，每3～7年，東太平洋赤道地區水溫會升高約攝氏2～3度，這個海水變溫暖的現象是聖嬰現象的特徵。聖嬰現象會造成全球降水區域分配的改變，南美會有大雨，澳洲、印尼、南亞則會有嚴重的乾旱發生。就較小的尺度來說，海水的溫度會影響熱帶氣旋（颶風、颱風）的生成發展，因為熱帶氣旋是借由暖水取得能量來發展其結構和強度的。\r\n\r\n熱帶氣旋的形成與維持，需要表層海水溫度高於攝氏26度。一般而言，SST越高，強度越強，其形成的機率也越高。然而，還有許多因素影響熱帶氣旋的強度。遙測SST數值能用在偵測颶風可能發生前的表層水溫特徵，一般來說，當颶風通過後，觀測到SST較低，因混合層深度加深、表面熱量散失。另外，海平面風場輻散造成的湧升流也使得SST較低。\r\n\r\nSST的改變和生物的多寡也有重要的關聯，這些生物包含：浮游生物、海草、貝類、魚類、哺乳類等。想知道台灣附近海域的海水表面溫度，可查閱中央氣象局網站－海象測報－海溫測報－海溫分析圖（臺灣）。想知道即時全球海水表面溫度，可上網查詢。\r\n\r\n臺北市立萬芳高級中學地球科學科邱怡禎老師/國立臺灣師範大學地球科學系吳朝榮教授責任編輯",
